Is it the route that follows Knot Gill up on the eastern side to Knot Farm, Then up to Pinhaw via Sunny Side? There is also a route on the western side of Knot Gill if not.

No, it starts at the east end of the village and goes up the bridleway running north before dropping down to Tow Top Farm then heads NW to join up with the Pennine Way. It then follows the PW to Pinhaw and returns via the same route.
I have never been on either of the two paths you mention but they appear to have quite a few stiles which isn't good for a race. I haven't seen the markers for the upper path on to Pinhaw - I'll have to check this out.
